紫外线B照射低温冷冻同种异体神经移植的实验研究 被引量:11

摘要 目的:观察低温冷冻加紫外线B照射的大白鼠的异体神经与新鲜自体神经移植的差异,探讨神经移植的再生机制。方法:以30只健康雄性大白鼠为实验对象,随机分成A,B,C3组,每组后肢一侧移植冷冻加紫外线B照射的同种异体坐骨神经,另一侧移植新鲜自体坐骨神经。于术后2,4,6个月,进行肉眼观察、电生理学检查、腓肠肌和比目鱼肌湿质量测定、组织学观察。结果:在术后2个月时,实验侧、对照侧功能均逐渐恢复,肌肉的颜色、体积两侧差异无显著性意义。诱发肌电图在2个月时呈多相性、低振幅波形,4~6个月时多相性波形减少,振幅增大,肌肉收缩力增强,诱发动作电位峰值,实验侧与对照侧差异无显著性意义;运动神经的神经肌肉传导速度、腓肠肌和比鱼肌湿质量测定实验侧与对照侧也差异无显著性意义(P>0.05);组织学观察两侧神经再生状态基本相似。结论:低温冷冻加紫外线B照射可以充分降低异体神经的免疫原性,与新鲜自体神经移植的效果相似,是具有应用前景的神经移植材料。 AIM:To observe the difference between frozen and ultraviolet B radiated nerve allografts and fresh nerve autografts,and explore the regeneration mechanism of nerve grafting. METHODS:Thirty healthy male rats were randomly divided into groups A,B and C,and then each group was divided into 2 subgroups.Sciatic allografts were grafted to right legs and fresh sciatic autografts were grafted to left legs respectively by freezing and ultraviolet B radiation in each group.The muscle color was observed by naked eyes.Electrophysiological examination,determination of wet masses of gastrocnemius muscle and soleus muscle,histological observation and t test were performed 2,4 and 6 months after operation. RESULTS:The functions of study and control sides were gradually recovered,and there were no obvious differences in the colour and size of muscles between the two sides two months after operation.At 2 months, the inducing electromyogram showed inhomogeneity and low amplitude waves.At 4 to 6 months,inhomogeneity waves were decreased,amplitude of vibration was enlarged and muscle contraction was strengthened.The frozen and ultraviolet ray B radiated nerve allografts were not different from fresh nerve autografts in the peak value of evoked action potential,muscle conduct velocity of motor nerve and the wet masses of gastrocnemius and soleus muscles(P >0.05). Histological observation showed that nerve regenerations were similar in both sides. CONCLUSION:The freezing and ultraviolet ray B radiation fully decreases the immunogenicity of nerve allografts,and the effect is similar to that of the fresh nerve autografts,it is a kind of nerve migrating material with application prospect.
