摘要
目的:研究抗抑郁药噻萘普汀能否影响吗啡处理大鼠脑内内源性焦虑物质-苯甲二氮结合抑制因子(diazepambindinginhibitor,DBI)mRNA的表达。方法:15只SD大鼠(四川大学动物实验中心提供),随机进入吗啡+生理盐水组、吗啡+噻萘普汀组和生理盐水组,每组5只。按照剂量递增每12h给大鼠腹腔注射盐酸吗啡(10~80mg/kg,共10次),每次注射吗啡前0.5h注射噻萘普汀(15mg/kg),对照组给予同体积的生理盐水,各组于末次吗啡注射后2h,给与腹腔注射盐酸纳洛酮2mg/kg,促发戒断,2h后处死大鼠,提取脑内总RNA,进行RT-PCR,检测DBImMRNA的表达。结果:吗啡+噻萘普汀组DBImRNA表达(0.52±0.08)明显少于吗啡+生理盐水组(0.91±0.08),差异有显著性意义q=15.631P<0.01);艹卓与生理盐水组(0.40±0.06)差异无显著性意义(q=1.992,P>0.05)。结论:噻萘普汀能够影响吗啡处理大鼠脑内DBImRNA的表达。
AIM: To investigate whether tianeptine can affect the mRNA expression of diazepam binding inhibitor(DBI) in the brain of morphine treated rats.METHODS:A total of 15 Sprague Dawley(SD) rats, provided from Animal Experimental Center of Sichuan University, were randomly divided into three groups: morphine &saline, morphine &tianeptine and saline with five in each group. Rats were intraperitoneally injected with morphine hydrochloride(10 to 80 mg/kg) increasingly per 12 hours totally for ten times. Tianeptine(15 mg/kg) was injected before half hour injection of morphine.The same volume of saline was treated in control group. After the last injection of morphine for two hours in each group, naloxone hydrochloride (2 mg/kg)was intraperitoneally injected in order to facilitate abstinence syndrome. Rats were sacrificed after two hours, and the total RNA in the brain was extracted to assay the expression of DBI mRNA with reverse transcriptase polymerase chain reaction(RT PCR).RESULTS: The expression of DBI mRNA(0.52±0.08) in morphine &tianeptine was less than that in morphine &saline group (0.91±0.08), which was significantly different(q=15.631, P< 0.01), and was not significantly different from that in saline group (0.40±0.06) (q=1.992, P >0.05).CONCLUSION: Tianeptine can affect the expression of DBI mRNA in the brain of rats treated with morphine.
