摘要
目的 以Wistar大鼠种植性肝癌模型为研究对象 ,探讨不同作用时间奥曲肽对肝部分切除术后肝癌细胞的凋亡变化。方法 4 8只雄性Wistar大鼠行肝左叶切除加肝右叶肿瘤种植后 ,随机分为对照组 (n =16 ) ,奥曲肽治疗组 (n =16 ) ,奥曲肽延迟治疗组 (n =16 )。术后 1周、2周各取 8只处死 ,运用碘化丙啶 (PI)染色法和荧光素标记的膜联蛋白V(FITC AnnexinV)联合碘化丙啶 (PI)染色法检测肝癌细胞凋亡。结果 术后 1周和 2周时 ,治疗组肿瘤细胞的凋亡率明显高于对照组 ,差异有显著性 (P <0 0 1) ,所不同的是 1周时 ,治疗组的细胞凋亡率高于延迟治疗组 (P <0 0 1) ,而 2周时二者间则无差异 (P >0 0 5 )。结论 奥曲肽可以有效诱导大鼠种植性肝癌细胞的凋亡 。
Objective To determine whether octreotide (OCT) can induce apoptosis of implanted liver cancer cells in rat model. Methods A total of 48 male Wistar rats weighing 180-220 g were randomized into 3 groups of OCT-12h group, OCT-72h group and control group with 16 in each. All rats had the left lobe of liver resected. Walker-256 tumor was implanted and SS intraperitoneally administered at 50 μg/kg into rats in the former groups. Flow cytometry was used to determine whether OCT can induce apoptosis of transplanted tumor cells in the 1st and 2nd week. Results The apoptotic rate of implanted tumor cells was significantly higher in the former 2 groups than in the control (P<0.01) in the 1st and 2nd week. However, the apoptotic rate was markedly higher in OCT-12h group than in OCT-72h group in the 1st week (P<0.01). This difference disappeared in the 2nd week (P>05). Conclusions OCT can induce apoptosis of implanted tumor cells in the rat model. Meanwhile, its inducing effect is time-dependent in the early stage.
