摘要
目的:研究胰岛素对减轻脑缺血再灌注损伤的作用机制。方法:制备肾血管性高血压大鼠(RHR),用线栓法复制大脑中动脉阻塞(MCAO),造成缺血2h后分别再罐注1,3,5d,并于再罐注后开始使用胰岛素,采用TUNEL法原位标记DNA片段,检测凋亡细胞的变化。结果:实验组TUNEL阳性细胞明显减少(P<0.05)。结论:胰岛素减轻脑缺血再灌注损伤的机制与减少再灌注后细胞凋亡或死亡有关。
Objective: To study the mechanism of insulin treatment on reperfusion injury after ischemia. Methods: The renovascular hypertension rat(RHR) were don ed to MCAO as described by Zea Longa for 2 hours and treated immediately with insulin after reperfulsion. The a poptotic cells were detected at 1,3,5 days after reperfusion with TUNEL method. Results: The numbers of the T UNEL positive cells were significantly lower in the tested groups than the controlled groups(P < 0 .05). Conclusion: Inhibitting apoptosis after reperfusion is probably one of the mechanisms that insulin reduc es reperfusion injury.
