摘要
目的:探讨通过理化方法处理制备的复合型完全脱蛋白骨(CFDB)的细胞相容性,为骨缺损修复寻找理想的组织工程支架材料。方法:将猪肋骨用理化方法制得CFDB,并进行CFDB的成分检测、孔隙结构观察及CFDB对人胎骨膜成骨细胞的形态特征、ALP活性、细胞周期、倍体水平的影响观察。再将其与成骨细胞体外复合培养,以了解其细胞相容性及细胞毒性。结果:CFDB的主要成分为羟基磷灰石,蛋白质含量几乎为零,仍具有原骨组织的网状孔隙结构系统,其原骨组织骨盐支架的三维多孔结构存在,并对人胎骨膜成骨细胞的形态特征、ALP活性、细胞周期、倍体水平无有害影响。结论:CFDB具有原骨组织的网状孔隙结构系统,具有良好的细胞相容性,可望用于成骨细胞的支架材料。
AIM:To study the cellular compatibility of composite fully deproteinised bone(CFDB) prepared with physical and chemical methods so as to look for the trestle materials of bone tissue engineering for the repair of bone defect. METHODS:Pig ribs were made into composite fully deproteinised bone with physical and chemical methods.The ingredient and natural pore system were observed. The effect of CFDB on cell feature, alkaline phosphatase(ALP) activity and cell cycle of human embryonic periosteal derived osteoblasts was also studied.Then the cellular compatibility and cytotoxicity were examined after combined culture of osteoblasts and CFDB in vitro. RESULTS:The main ingredient of CFDB was hydroxyapatite,it hardly had any protein, but it had natural pore system,which possessed pore network system of original bone.It had no bad effect on cell feature,ALP activity and cell cycle of human embryonic periosteal derived osteoblasts composited with CFDB. CONCLUSION:CFDB possesses the natural pore system of original bone and the good cellular compatibility,and it can be used as the trestle materials of osteoblasts.
