摘要
目的:探讨姜黄素对脑出血大鼠脑组织肿瘤坏死因子(tumornecrosisfactor-alpha,TNF-α)水平的影响。方法:将120只健康雄性SD大鼠随机分为姜黄素治疗组、脑出血对照组和假手术组。每组分为手术前、术后6,12,24,48,72,96,120h8个时点,每个时点5只大鼠。采用立体定向自体血注入大鼠尾状核建立脑出血模型,观察各组术前及术后各时点血肿周围脑组织肿瘤坏死因子(TNF-α)水平及含水量变化。结果:脑出血对照组术后12~96h各时点血肿周围脑组织TNF-α水平犤分别为(1.48±0.13),(1.53±0.20),(1.56±0.39),(1.49±0.28),(1.47±0.21)μg/L犦较假手术组犤分别为(1.06±0.33),(1.05±0.28),(1.14±0.15),(1.04±0.14),(1.08±0.11)μg/L犦明显升高(P<0.05)。姜黄素治疗组术后12~96h各时点TNF-α水平犤分别为(1.12±0.21),(1.11±0.18),(1.21±0.11),(1.15±0.15),(1.14±0.21)μg/L犦均明显低于脑出血对照组(P<0.05)。脑出血对照组和姜黄素治疗组术后各时点血肿周围脑组织含水量分别与相应脑组织TNF-α水平呈正相关(r=0.541,P<0.05;r=0.548,P<0.05)。结论:姜黄素可抑制大鼠脑出血后TNF-α含量增加,其对抗脑出血后脑水肿作用可能与此有关。
AIM: To explore the influence of curcumin on the level of tumor necrosis factor-alpha (TNF-α) in the peri-hematoma brain tissue of rats after intracerebral hemorrhage (ICH). METHODS: A total of 120 healthy male SD rats were assigned randomly into curcumin treated group, ICH control group and sham-operated group, and each group was subdivided into 8 various time points: before, 6, 12, 24, 48, 72, 96 and 120 hours after operation, with 5 rats in each subgroup. Rat models of ICH were established by injecting autologous blood into the caudate nucleus. The changes of TNF-αlevels and water content in the peri-hematoma brain tissue at various time points before and after operation were analyzed. RESUITS: The levels of TNF-αat various time points from 12 to 96 hours after operation in the ICH control group [(1.48±0.13), (1.53±0.20),(1.56±0.39),(1.49±0.28) and (1.47±0.21) μg/L] were obviously increased as compared with those in the sham-operated group[(1.06±0.33),(1.05±0.28),(1.14±0.15),(1.04±0.14) and(1.08±0.11)μg/L](P< 0.05); while those in the curcumin treated group [(1.12±0.21), (1.11±0.18),(1.21±0.11),(1.15±0.15) and(1.14±0.21) μg/L]were significantly lower than those in the ICH group(P< 0.05). The water content in the peri-hematoma brain tissue at various time points after operation were significantly positively correlated with the levels of TNF-αin the corresponding brain tissue of ICH control group and curcumin treated group(r=0.541,P< 0.05;r=0.548,P< 0.05). CONCLUSION: Curcumin can inhibit the increase of TNF-αlevel in rat after ICH, which may be related with its anti-edema effect following ICH.
