摘要
背景:脑缺血、癫痫等损伤可刺激成年海马的齿状回颗粒细胞下层(subgranularzone,SGZ)神经干细胞增殖分化。去大脑皮质血管状态下引起碱性成纤维细胞生长因子(basicfibroblastgrowthfactor,bFGF)表达增多是否是刺激SGZ神经干细胞增殖的主要原因之一呢?目的:探讨去皮质血管对成年大鼠海马齿状回SGZ神经干细胞增殖、分化的影响以及去大脑皮质血管状态下bFGF的表达情况。设计:随机对照实验研究。地点和对象:实验地点:北京中医药大学基础医学院科学试验中心。成年Wistar雄性大鼠33只,随机分为正常组(15只)和模型组(18只)。干预:采用去Wister大鼠脑皮质血管脑缺血模型及免疫组织化学方法观测大鼠海马齿状回溴化脱氧尿核苷(bromodeoxyuridine,BrdU),Tuj-1,Vimentin,Nestin,增殖细胞核抗原(proliferationneuclearantigen,PCNA),bFGF的表达情况。主要观察指标:大鼠海马齿状回的BrdU,Tuj-1,Vimentin,Nestin,PCNA,bFGF的表达情况。结果:去大脑皮质血管后成年大鼠SGZ中BrdU,Tuj-1,PCNA阳性细胞的数目增多。模型组大鼠齿状回BrdU免疫阳性细胞数目犤去在脑皮质15d为(46.667±6.614)个/脑片,去大脑皮质30d为(14.111±3.408)个/脑片〗与正常组犤9.556±1.236)个/脑片犦比较,差异有显著性意义(P<0.01)。
BACKGROUND:Proliferation and differentiation of neural stem cells(NSCs) is found increased at subgranular zone(SGZ) in hippocampal dentate gyrus of adult rat during brain ischemia,epilepsy or other impairments, whether it is mainly due to the increased expression of basic fibroblast growth factor(bFGF) under cortical devascularization state is poorly acknowledged. OBJECTIVE:To investigate the changes of proliferation and differentiation of NSCs in hippocampal dentate gyrus of adult rats after cortical devascularization,as well as the expression of bFGF. DESIGN:A randomly case control study. SETTING and MATERIALS:This study was carried out at the Experimental Center of the College of Basic Medicine, Bejing University of Chinese Medicine.Thirty-three adult male Wistar rats were randomly divided into normal group(n=15) and model group(n=18). INTERVENTIONS:Cortical devascularization brain ischemic model was established in Wistar rat for observing the expression of bromodeoxyuridine(BrdU),Tuj-1,vimentin,nestin,proliferation nuclear antigen(PCNA) and bFGF in dentate gyrus by immunohistochemical assay. MAIN OUTCOME MEASURES:The expression of BrdU,Tuj-1, vimentin,nestin,PCNA and bFGF in dentate gyrus. CONCLUSION:Self-regeneration and multiple differentiation capability of NSCs in hippocamps SGZ can be enhanced through up-regulation of bFGF and subsequently connection with receptors in the manner of autocrine or paracrine due to brai
