摘要
目的研究Wnt-1基因在全反式维甲酸(ATRA)诱导人胚胎神经干细胞分化为神经元过程中的表达及其作用。方法用1μmol/L的维甲酸诱导人胚胎神经干细胞,诱导7d后,做NSE免疫荧光染色,计数分化为神经元的比例。于ATRA诱导前、诱导3d和诱导7d,提取细胞的总RNA。用半定量RT-PCR法检测Wnt-1基因的表达情况。结果与对照组相比,全反式维甲酸能显著提高人胚胎神经干细胞分化为神经元的比例(P<0.01)。Wnt-1基因在维甲酸诱导后明显上调(P<0.001)。结论Wnt-1基因在全反式维甲酸诱导人胚胎神经干细胞分化为神经元的过程中起正调控作用,Wnt-1基因与神经元的分化关系密切。
Objective To investigate the expression and effect of Wnt-1 gene in the course of the human embryonic neural stem cells induced by all-trans-retinoic-acid(ATRA) differentiating into neurons. Methods The human embryonic neural stem cells were induced differentiation with 1μmol/L ATRA in vitro. The immunocytofluorescence was performanced after seven days; quantitation of the neurons was obtained by counting NSE positive cells. Total RNA was extracted from the cells that before inducing, inducing 3 day, inducing 7 day. The expression of Wnt-1 gene was tested by semi-quantitative RT-PCR assay. Results The inducing differentiated rates of neurons were improved greatly by ATRA compared with control(P<0.01). The expression of Wnt-1 gene increased significantly after ATRA treated(P<0.001). Conclusions There are the important role for the expression of Wnt-1 gene during the human embryonic neural stem cells differentiated into neurons, the gene plays the positive-regulation role.
