摘要
黑曲霉 (Aspergillusniger)A3经离子注入后选育出高产木聚糖酶突变株AN497,通过摇瓶液态发酵产酶培养基的L16 ( 4 4)正交实验 ,得到了优化的培养基配方 (g/L) :玉米芯粉 80 ,蔗糖 6,复合氮源 (NH4 )SO4 +NaNO3(质量比为 1∶2 ) 1 2 ,吐温 2 0 3 ,用无氮的Mandels氏无机盐溶液配制。同时对产酶菌株的发酵过程进行了试验 ,发酵 84h达产酶高峰 ,木聚糖酶活力可达 646IU/mL ,比培养基未优化前 ( 5 84 8IU/mL)提高了 1 0 5 %。
AN497, a high-yielding xylanase strain, was screened by the technology of ion implantation. Through orthogonal experiment,the optimal condition was obtained for producing xylanases of Aspergillus niger AN497.The suitable medium contained(g/L): corn-core 80,cane sugar 6, complex nitragen resource(NH 4)SO 4+NaNO 3 (mass ratio 1∶2)12, Tween20 3, prepared in Mandels nutritional solution. Xylanase producing process by strain AN497 was studied, and the result showed the optimum time for xylanase secretion was about 84 ?h, the xylanase activity is as high as 646IU/mL. Xylanase activity was increased by 10. 5% compared to that of former one strain M-2980.
