The Effect of Age on Proliferation of Human Mesenchymal Stem Cells in Vitro

摘要 目的 观察不同年龄段人骨髓间充质干细胞 (humanmesenchymalstemcells ,hMSCs)体外生长、增殖的影响。方法 使用密度梯度离心法分离不同年龄人骨髓MSCs进行培养 ,保留贴壁细胞传代 ,观察细胞生长情况 ,检测其增殖活性、细胞周期。结果 低龄hMSCs较高龄hMSCs体外生长快、MTT活性高以及增殖期比例大。结论 hMSCs的增殖能力和活性随着年龄的增加而降低 ,建立其增殖参数可满足组织工程种子细胞在临床应用中不同患者的要求。 Objective To investigate the effects of the donor age on the growth of human mesenchymal stem cells(hMSCs) in vitro. Methods HMSCs were isolated by using density gradient centrifuge method from 39 donors of different ages (7 to 79 years old), and cultured in vitro. The adhesive cells were preserved to passage, then their growth and proliferation capability were determined. Results The hMSCs from younger donors showed a stronger capability of proliferation compared with the hMSCs from older donors. Conclusion The proliferation capability of hMSCs decreased with aging. The results have significant implications for the proposed clinical application of hMSCs.
