摘要
目的 探讨平阳霉素 (PYM )对体外培养的喉癌细胞凋亡的作用 ,并研究其作用的量 -效关系。方法 取人喉鳞状细胞癌株Hep - 2细胞进行细胞培养 ,PYM作用不同浓度不同时间后 ,以TUNEL染色、电镜、流式细胞仪检测凋亡情况。结果 高浓度 (10 0 ,10 0 0 μg/ml)PYM作用下 ,靶细胞呈典型凋亡改变。低浓度 (1,10 μg/ml)PYM作用下未见明显凋亡改变。在一定范围内 ,PYM诱导细胞凋亡作用呈浓度、时间依赖性。 结论PYM可诱导人喉癌Hep - 2细胞出现凋亡。PYM诱导细胞凋亡浓度较高 ,诱导喉癌细胞凋亡可能是PYM化疗作用的重要机制之一。
Objective To demonstrate whether Pingyangmycin can induce the apoptosis in human larynx squamous cell line (Hep-2). Methods Hep-2 cells were exposed to different concentrations of PYM and evaluated by TUNEL and Electron microscope. Flow cytometric DNA analysis was used to analyze the peak appeared in DNA frequency distribution histograms.Results Hep-2 cells in the presence of low concentrations of PYM (1μg/ml, 10μg/ml) showed no significant apoptosis. In contrast, cells in the presence of high concentration (100μg /ml, 1000μg /ml) exhibited the morphological and biochemical changes associated with apoptosis.To some extent,the effect was dose-time dependent.Conclusions The results of study demonstrate that Pingyangmycin can induce Hep-2 cells apoptosis,which has the clinical significance of the chemotherapy of human larynx squamous cancer cells.
