摘要
目的 研究免疫组化技术中 3种抗原加热修复方法的效果。方法 用免疫组化SABC法检测五种单克隆抗体 ,分别使用水煮法、微波法、高压法修复抗原 ,从中选出最佳修复方法。结果 单克隆抗体BCL - 2 ,BAX ,P5 3,BDNF ,VEGF在 2种修复液中显色结果相同或相似 ,而高压法明显优于微波法及水煮法 ;P5 3在Tris-EDTA (pH9 0 )修复液中效果明显优于柠檬酸盐缓冲液 (pH6 0 )。 结论 Tris -EDTA (pH9 0 )高压法的实验重现性与染色强度均优于微波法及水煮法。
Objective To study the effects of three methods of heating induced epitope retrieval in immunohistochemistry. Methods Check five kinds of antibody with immunohistochemistry SABC method and chose the best antigen retrieval method among poach method, microwave method and high pressure method respectively. Results Although the same or similar result of color appearance is shown between two retrieval liquids when putting BCL-2,P53,BDNF into them,the high pressure method is better than the microwave method and poach method. The effect of P53 in Tris-EDTA (pH9.0)is better than in citrate amortize liquid. Conclusions The recurrence and intension of appear color of high pressure method with Tris-EDTA amortize liquid are better than that of microwave method and poach method.
