摘要
目的:通过观察亚低温对缺血再灌注损伤大鼠视网膜组织型纤溶酶原激活物(tissue-typeplasminogenactivator,TPA)含量的影响,探讨亚低温防治缺血再灌注视网膜损伤的机制。方法:选择健康成年雄性SD大鼠,随机分成正常对照组、缺血再灌注组和亚低温缺血再灌注组。采用提高眼压法造成视网膜缺血后,恢复眼压形成血流再灌注。于缺血1h再灌注1h、缺血1h再灌注2h、缺血2h再灌注1h、缺血2h再灌注2h时测定各组大鼠视网膜组织TPA的含量。结果:缺血再灌注组的大鼠视网膜组织TPA含量高于正常对照组,亚低温缺血再灌注组的大鼠视网膜组织TPA含量低于缺血再灌注组。结论:缺血再灌注视网膜损伤与视网膜组织TPA含量升高有关,亚低温可通过降低视网膜TPA含量而减轻缺血再灌注视网膜损伤。
AIM:To explore the protecting mechanism of injury on retina induced by ischemia reperfusion(IR) under subhypothermia by observing the effect of mild hypothermia on the content of tissue type plasminogen activator(TPA) of retina in rats with IR injury. METHODS:Healthy adult male SD rats were selected and divided into normal control group,IR group and IR group with mild hypothermia.The intraocular pressure(IOP) method was used to occlude retinal blood vessels to form retinal ischemia,afterwards IOP was lowered to normal level to allow reperfusion.The content of TPA in the retina of all rats were quantified after 1 hour ischemia and 1 hour reperfusion,1 hour ischemia and 2 hour reperfusion,2 hour ischemia and 1 hour reperfusion,2 hour ischemia and 2 hour reperfusion. RESULTS:The content of TPA in the retina of IR group was higher than that of normal control group.The content of TPA in the retina of IR group with subhypothermia was lower than that of IR group. CONCLUSION:The retina injury induced by IR is related with the increasing content of TPA,mild hypothermia can protect the retina induced by IR injury by decreasing the content of TPA.
