小鼠成骨细胞改良酶的消化分离培养 被引量:13

摘要 为研究细胞毒性物质对成骨细胞的影响,采用 24 h 内乳鼠头盖骨,对国外现行报道方法进行了改进,建立了适合我国同类试验室条件下的骨组织细胞培养方法——酶消化分离培养法,并用该新方法培养出了具有典型特征的成骨细胞系。研究结果表明,此法具有耗时短、消化充分和对细胞损伤小的特点,能够满足试验的要求。镜下细胞形态不规则,多呈三角形、多角形,有突起,胞质丰富、清晰。应用特异性染色方法,胞质中呈现相应的特征性染色。培养过程中,成骨细胞经历静止期、指数成长期和滞后期等三个阶段。培养 14 d 后,出现矿化结节。 The typical cell line was obtained through modified culture method to study the effects of cytotoxic substance on osteoblasts by enzyme digestion from calvaria of fetal rats within 24 hs. Microscopically, osteoblasts had irregular shapes. The triangular, polyangular shape and cell processes were observed in the majority of osteoblasts. Osteoblasts presented rich and clear cytoplasm. Characteristic stain in cytoplasm was seen with typical staining (Gomori Ca-Co staining). The osteobalsts showed lag period, the period of exponential growth, and the period of reduced or zero growth. Mineralized nodes were found at 14 days of culture. This study established a basic data for future research about the effect of fluorine on bone tissue.
