摘要
运用硝酸钬溶液对蚕豆根尖染毒,提取根尖细胞DNA进行琼脂糖凝胶电泳;取根尖压片观察细胞核异常情况;同时取根尖细胞进行单细胞凝胶电泳。结果表明,钬离子诱导了根尖细胞核异常;在8mg·L-1剂量下,彗星细胞尾长和拖尾细胞数量在24h内随着根尖处理时间的延长而增加,处理24h时,琼脂糖凝胶电泳中出现明显的连续拖尾带型;处理48h后彗星拖尾缩短,琼脂糖凝胶电泳中拖尾带型缩短,推测可能发生了DNA修复作用或者交联作用。64mg·L-1剂量处理24h,彗尾缩短且出现核碎裂,琼脂糖凝胶电泳中未出现拖尾带型。实验中未观察到细胞凋亡的典型DNA"梯状带型"和特征性彗星拖尾现象。
Soaked in different concentrations of holmium nitrate solution for different time, root tips of Vicia faba were washed and fetched. DNA of the root tips was extracted and performed agarose gel electrophoresis. Slides of the root-tip cells were prepared and examined under microscope for anomalous nuclei. Meanwhile, root-tip cells were prepared and performed single cell gel electrophoresis. The results show that anomaly of nuclei is induces by holmium ions. The reslts also show that comet tails are lengthened and numbers of tailed cells are on the increases with the extension of disposal within 24 h at concentration of 8 mg·L^(-1). and that at disposal of 24 h, an obvious and continual tape appeares in agarose gel electrophoresis, but at disposal of 48 h, the comet tails are shortened and only a short tailing tape shows in agarose gel electrophoresis. It may be the result of DNA repair or DNA cross-link. At concentration of 64 mg·L^(-1) for disposal of 24 h, the comet tails are shorter than those at 8 mg·L^(-1) for the same time. Furthermore, fractures of nuclei were observed under microscope, but no tailing tape was found in agarose gel electrophoresis. Neither typical DNA ladder, nor characteristic comet tails′ shape of apoptosis was found in the experiments.
