摘要
目的:研究中药灯盏花提取物对大鼠心脏移植排斥反应的作用。方法:以Wistar大鼠为供体,SD大鼠为受体,建立大鼠腹腔异位心脏移植模型,分为灯盏花提取物低剂量组(5mg·kg-1·d-1,E1)、高剂量组(10mg·kg-1·d-1,E2)、环孢素A(CsA)组和对照组(C组)。移植术后各实验组受体腹腔注射药物至移植心停跳。观察移植心存活时间、病理变化及受体外周血液中白细胞介素-2(interleukin-2,IL-2)水平。结果:E1组、E2组和CsA组受体大鼠外周血IL-2的水平明显较C组低,同时移植心脏的存活时间较C组延长,排斥反应减弱。结论:灯盏花提取物预处理可明显减轻大鼠移植心脏的排斥反应,并延长移植心脏的存活时间。
Objective: To study the anti-rejection effect of the extract of erigeron in heart transplantation rats. Methods: All groups were consisted of Wistar as donors and SD recipients, including low(5 mg·kg-1·d-1)and large(10 ng)doses of the extract of erigerion groups (E1 and E2), CsA group (E3), and control group(C). Recipients were given drugs one day after operation, and their blood samples were taken on the fifth postoperation day for the measurement of IL-2. Grafts survival days and the beating conditions of the heart were observed everyday. Results: The grafts survival days of E1, E2 and E3 groups increased significantly(P<0.05), the serum IL-2 of the above three groups decreased greatly(P<0.05), and the grafts' pathologic damages of them were slight compared with that of C group. Conclusions: The pretreatment by using the extract of the erigeron could probably reduce the pathologic damage of the graft and prolong the survival days.
