尼古丁对帕金森病大鼠纹状体脑胶质细胞源性神经营养因子和多巴胺含量的影响(英文)

摘要 背景:流行病学研究显示,吸烟和帕金森病(Parkinson'sdisease,PD)存在负相关系,吸烟可能对PD具有保护作用。人们一直在探索尼古丁对PD的保护作用机制。目的:研究尼古丁对PD大鼠纹状体脑胶质源性神经营养因子(glialcelltinederivedneurotrophicfactor,GDNF)和多巴胺含量的影响。设计:随机对照研究。地点和对象:实验地点:华中科技大学同济医学院协和医院,80只大鼠被随机分为预防组50只和治疗组30只。干预:本文第一作者将6-羟多巴胺(6-hydroxydopamine,6-OHDA)立体定向注射到大鼠右侧中脑腹侧被盖部和黑质致密部,建立大鼠模型。采用生化,免疫组织化学方法观察不同剂量尼古丁对PD大鼠的作用。主要观察指标:检测纹状体GDNF表达及多巴胺含量的变化。结果:造模前及造模后皮下注射尼古丁的PD大鼠,纹状体GDNF表达及多巴胺含量较PD组有明显改善(P<0.05)。结论:尼古丁可减轻6-OHDA对黑质多巴胺能神经元的损伤,对PD大鼠具有保护作用。 BACKGROUND:The epidemiological study indicates that there is negative correlat ion between smoking and Parkinson's disease(PD),which means that smoking might h as protective reaction to PD. Therefore,many efforts were inserted into the rese arches on the protective mechanism of Nicotine on PD. OBJECTIVE:To study the impact of nicotine on the contents of glial cell line- derived neurotrophic factor(GDNF) and dopamine in corpus striatum of PD rats. DESIGN:A random control study. SETTINGS and PARTICIPANTS:Study setting:Study was completed in Xiehe Hospital affiliated with Tongji Medical Faculty of Huazhong Science and Technology Univer sity.80 rats were randomly allocated into prevention group(n=50) and treatment g roup(n=30). INTERVENTIONS:The rat model was established by the first author through solid directional injection of 6-hydroxydopamine(6-OHDA) into ventral tegmentam(VTA) and pars compata of substantia nigra compact part(SNpc) of midbrain on the righ t side in rats.To observe the impact of different dose nicotine on the PD rats t hrough biochemical immune constitution chemical measure. MAIN OUTCOME MEASURES:The expression of GDNF and the dopamine content in corpu s striatum were detected. RESULTS:The expression of GDNF and dopamine content in corpus striatum in the PD rats before and after modelling by subcutaneous injection of nicotine signifi cantly improved compared with that of PD group(P< 0.05). CONCLUSION:Nicotine could alleviate the injury of 6-OHDA on dopaminergic neur ons in substantia nigra,which provide protection on PD rats.
