Protective Effect of Supernatant of Lymphocytes Modified by Brain-derived Neurotrophic Factor Gene on PC12 Cells Damage Induced by Oxidative Stress

摘要 【目的】研究脑源性神经营养因子基因(brain-derivedneurotrophicfactor,BDNFgene)修饰淋巴细胞对PC12细胞氧化应激损伤的保护作用。【方法】MTT法检测H2O2和多巴胺(dopamine,DA)对PC12细胞生长的抑制率,Hoechst染色和PI染色流式细胞仪(flowcytometry,FCM)检测H2O2和DA对PC12细胞凋亡的诱导作用。【结果】BDNF基因修饰的大鼠淋巴细胞的培养上清可降低H2O2和DA对PC12细胞生长的抑制率并能抑制H2O2和DA诱导PC12细胞凋亡。【结论】BDNF基因修饰淋巴细胞的培养上清对PC12细胞氧化应激损伤具有保护作用。 ObjectiveTo study the protective effect of lymphocytes modified by brain-derived neurotrophic factor gene (BDNF gene) on PC12 cells damage induced by oxidative stress. MethodsThe growth inhibitory rate of PC12 cells mediated by H2O2 and dopamine (DA) was evaluated by MTT assay. The apoptosis of PC12 cells induced by H2O2 and DA was determined by PI stain flow cytometry and Hoechst stain. ResultsThe supernatant of lymphocytes modified by BDNF gene decreased the growth inhibitory rate and apoptosis rate of PC12 cells induced by H2O2 and DA. ConclusionThe supernatant of rat lymphocytes genetically modified by BDNF gene has the protective effect on PC12 cells damage induced by oxidative stress.
