摘要 【目的】检测退变椎间盘髓核组织中MMP-2的表达,进一步阐明椎间盘退变的机制。【方法】利用RT-PCR、Westernblot技术和明胶酶谱法对45例退变椎间盘髓核、12例非突出椎间盘髓核的MMP-2进行了检测。【结果】正常髓核内有一定量的MMP-2表达,退变髓核MMP-2mRNA、蛋白质表达水平和酶活性明显升高,分别是正常组的2.30,2.01和1.75倍,差异显著(P<0.01,P<0.01,P<0.01)。【结论】MMP-2和髓核退变关系密切。 ObjectiveThe expression of matrix metalloproteinase-2 (MMP-2) was investigated to understand the mechanism of intervertebral disc degeneration. MethodsTotal RNA and protein were extracted from 45 degenerative intervertebral disc nuclei and 12 normal ones. The expression of MMP-2 was investigated with reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analysis, respectively, and its activity was evaluated by gelatin zymography. ResultsRT-PCR and Western blot analysis indicated that expression of MMP-2 was present in normal samples. Compared with the normal nuclei, the expression of MMP-2 mRNA, protein and its activity in the degeneration nuclei were increased obviously by 130.58%(P< 0.01), 101.22%(P< 0.01), and 75.42%(P< 0.01), respectively. ConclusionMMP-2 is assumed to play a pivotal role in disc tissue degeneration.
