摘要
目的 研究SmB/B’抗原表位的基因免疫诱导自身抗体的产生的可能性。方法 构建SmB/B’抗原表位的真核表达载体,利用基因免疫的方法免疫同系Balb/c小鼠,用免疫印迹法测定抗SmB/B’抗体及多谱系抗体,用免疫荧光法检测抗核抗体核型。结果SmB/B'自身抗原表位为基础的基因免疫正常小鼠诱导了多种抗核抗体的产生,除抗SmB/B’抗体外,还包括抗SS-A、SS-B、rRNP、Scl-70,这些抗体可与细胞的多种成分相结合。结论 抗SmB/B’抗体可由自身SmB/B’的抗原表位诱生,并可进一步诱导多种抗核抗体的生成。
Purpose: To investigate whether gene immunization based on the SmB/B' epitope can induce the production of autoantibody. Methods: We constructed the eukaryotic expression vector-pcDNA3fSmB/B' which contained the target DNA fragment encoding epitope of SmB/B'. Then we injected the syngeneic Balb/c mice with the plasmid DNA intramuscularly. The antibodies against SmB/B' and polyspectrum antibodies were determined by immunoblot assay. The patterns of antinuclear antibodies were observed by immunofluorescent stain. Results: Many kinds of antinuclear antibodies were induced by gene immunization based on the major epitope of SmB/B', and these autoantibodies could bind to many kinds of components of the cells. As we expected, none of the mice in control group produced ANAs. Conclusions: Anti-SmB/ B' antoantibody can be induced by self epitope of SmB/B', and many kinds of other autoantibodies can produced gradually.
