摘要
本研究将酶标记葡萄球菌A蛋白(PPA)这一通用试剂引入间接夹心酶联免疫吸附试验(ELISA),建立于夹心PPA—ELISA,用于检测猪轮状病毒时显示出优越性,比夹心ELISA敏感,背景染色极低,健猪粪便的背景光密度与PBS相同。
This sandwich PPA-ELISA which incorporates HRP-SPA with normal sandwich ELISA has been newly developed for the detection of porcine rotaviruses. The reagents used are: hen anti-MA 86 IgG for caoting the wells; guinea pig anti-MA86 sera as the detecting antibody and HRP-SPA as the enzyme labelled probe. The specificity and sensitivity are proved satisfactory because the method shows little or no background when the specimens from healthy control piglets are detected and because optical density more than 1- 5 can be got when the specimens from infected piglets, which only give optical density lower than 0. 5 by the normal ELISA, are detected. So it may become a more useful method than the normal ELISA, for it is very easy to get result with the naked eye.
