摘要
目的:研究蛋白激酶A和蛋白激酶C在肾阳虚发病中的作用及补肾药对其调整作用。方法:用长期大量糖皮质激素造成肾阳虚大鼠模型,用32P-ATP参入反应底物中的摩尔数表示蛋白激酶的活性,用放免法测定cAMP,cGMP含量。结果:肾阳虚大鼠血浆中cAMP水平下降,cGMP水平升高,cAMP/cGMP比值降低,下丘脑组织中PKA和PKC活性在胞液和胞膜中的活性较正常组明显降低。补肾方药对PKA和PKC的活性均有不同程度的调整作用。结论:PKA和PKC可能参与了肾阳虚大鼠的发病机制,补肾方药可部分调整PKA和PKC的活性,从而改善肾阳虚症状。
Objective:Study the changes of the levels of protein kinase A and protein kinase C in kidney yang deficient rat and the regulation of tonifying kidney recipes.Methods:The experimental model of the kidney yang deficient rat was established by injecting the rats with cortison acetatis.The cAMP ands cGMP levels were determined with radioimmunoassay analysis.PKA and PKC activities were assessed by the incorporation of 32P from radiolabeled ATP into histone in the presence or absence of stimulators.Results:The levels of cAMP decreased,the levels of cGMP increased and the ratio of cAMP/cGMP decreased in kidney yang deficient rat plasma,compared to the normal rats.The cytosolic and membrane PKA and PKC activities in kidney yang deficient rat hypothalamus decreased significantly,compared to the control.The tonifying kidney recipes modulated the PKA and PKC activities in different degree.Conclusion:PKA and PKC may be responsible for the mechanism of kidney yang deficient symptom.The tonifying kidney recipe modulated at least in part the activities of PKA and PKC,and then improved in symptom of kidney yang deficiency.
