摘要
介绍了冷冻蚀刻技术的基本原理、方法和制得满意生物样品的技术关键。该技术是研究生物膜大分子结构的重要方法。使用HUS-5GB真空镀膜仪及冷冻复型装置HFZ-1和冷冻蚀刻装置FE-1两个附件实现冷冻蚀刻技术。经过对生物样品的快速冷冻、腐蚀、复型膜的清洗和捞膜方法等的改进,成功率达90%以上,可得到结构清晰,分辨率高的冷冻蚀刻图像。
The principle and methods of satisfactory freezing-etching are introduced. The
freezing-etching was carried out by using HUS-5 and HUS-5GB vacuum evaporators and two
accessories Model HFZ-1 specimen freeze-replica instrument and Model FE-1 freezing-etching
equipment. The success rates exceed 90% and the freezing-etching images have a clear structure
and high resolution after improving the methods of quick freezing for biological samples and of
etching and washing specimen replicas and of draging a film.
