摘要 目的 :观察糖基化终末产物 (AGEs)对大鼠肾小球系膜细胞基质金属蛋白酶 2 (MMP- 2 )活性和表达的影响。方法 :体外培养正常大鼠肾小球系膜细胞 ,糖化牛血清白蛋白作为刺激物 ,未经糖化的牛血清白蛋白及常规培养作为对照 ,分别用含 10 0 m g· L- 1 AGEs (AGEs组 )、 10 0 mg· L- 1 牛血清白蛋白 (BSA组 )和不含刺激物 (DMEM组 )的无血清 DMEM培养基 ,作用于系膜细胞 4 8h后 ,酶谱法检测各组系膜细胞上清 MMP- 2活性 ,逆转录聚合酶链式反应 (RT- PCR)检测系膜细胞 MMP- 2 m RNA的表达。结果 :与 BSA组及 DMEM组比较 ,AGEs组系膜细胞 MMP- 2 m RNA的表达明显下降 (P<0 .0 1) ,上清 MMP- 2活性明显降低 (P<0 .0 1)。结论 :AGEs能降低大鼠肾小球系膜细胞 MMP- 2的活性和表达。 Objective To investigate the effects of advanced glycation end products (AGEs) on matrix metalloproteinase-2 (MMP-2) activity and expression in rat mesangial cells. Methods Rat mesangial cells were treated with 100 mg·L -1 AGE-modified bovine serum albumin(AGEs group)or native bovine serum albumin(BSA group)for 48 h. Normal mesangial cells without any treatment were as a control (DMEM group). MMP-2 activity and mRNA were analyzed by zymography analysis and reverse transcriptase polymerase chain reaction (RT-PCR), respectively. Results Both MMP-2 activity and mRNA expression of mesangial cells in AGEs group were reduced significantly, compared with BSA or DMEM group (P<0.01). Conclusion AGEs can reduce the activity and expression of MMP-2 in rat mesangial cells.
