摘要
目的 研究雷公藤内酯醇(TRI)对缺氧再给氧大鼠心脏微血管内皮细胞和中性粒细胞黏附的影响。方法 培养分离大鼠心脏微血管内皮细胞,建立内皮细胞缺氧再给氧模型,分离大鼠的中性粒细胞,通过凝胶电泳迁移率(EMSA)测定核转录因子-κB(NF-κB)的活性,内皮细胞和中性粒细胞的黏附模型测定内皮细胞和中性粒细胞的黏附率。结果 大鼠心脏微血管内皮细胞在缺氧刺激后内皮细胞和中性粒细胞的黏附增加,NF-κB活性明显增高,再给氧后升高更明显;TRI组内皮细胞NF-κB活性明显降低(P<0.01),内皮细胞和中性粒细胞的粘附率显著下降(P<0.01),且呈剂量依赖效应。结论 TRI能明显抑制缺氧再给氧大鼠心脏微血管内皮细胞NR-κB活性,抑制内皮细胞和中性粒细胞的粘附。
Objective To study effects of Triptolide on adherence of neutrophil and rat cardiac microvascular endothelial cells (RCMECs) . Methods RCMECs were cultured and hypoxia-reoxgena-tion model of endothelial cells were developed. Neutrophil were isolated from blood, adhesion rate of endothelial cells and neutrophil were determined by the adhesion model. NF-κB activity of endothelial cells was determined by electrophoretic mobility shift assays (EMSA). Results NF-κB activity, adherence rate of neutrophil and RCMECs treated with hypoxia were increased markedly, and after re-oxgenation, these changes were more obvious. When endothelial cells were treated by reoxgenation supplemented with Triptolide, adhesion rate of endothelial cells was decreased markely in a dose-dependent manner; NF-κB activity was decreased apparently compared with H/R(P<0.01). Conclusion Triptolide inhibits adhesion rate of RCMECs and neutrophil mediated apparently by NF-κB.
