摘要
以南美水仙的鳞茎、叶片和叶柄等营养器官作为外植体进行组织培养 ,着重研究南美水仙最佳外植体、最佳培养基配方的关键技术。研究结果表明 ,诱导的最佳培养基为 MS+6 - BA2 .0 mg/ L+NAA3.0 mg/ L,培养 4 5 d可获得较多的小鳞茎。外植体分化能力的研究表明 ,南美水仙的不同类型外植体能力大小为 :鳞茎 >叶柄 >叶片 ;从鳞茎的不同部位来看 ,基部鳞茎盘 >内层鳞片下部 >外层鳞片下部 >内层鳞片上部 >外层鳞片上部。
The bulbs, leaves and leafstalks of Eucharis grandiflora were used as explants in study of organ culture to establish a rapid technique system of propagation in vitro, and to find out the best explants, medium culture procedure. The results are as follows: A best medium of inducement were MS+6-BA 2.0 mg/L+NAA 3.0 mg/L, A good many to small bulbs can be got after 45 days culture. Effects of different explants on shoot regeneration: bulbs> leafstalks > leaves. Effects of different parts bulbs on shoot regeneration: basilar tray of bulbs>infrastructure of endothecium’s squama> infrastructure of outboard squama > top of endothecium’s squamas > top of outboard squama.
