摘要 【目的】研究高碘诱导脑细胞凋亡的机制。【方法】以高碘水喂养豚鼠 180d复制出高碘甲状腺肿模型 ,应用流式细胞术及免疫荧光染色的方法检测高碘对大脑皮质、海马组织细胞凋亡及细胞周期的影响 ,Bcl 2、Bax基因蛋白表达与细胞凋亡的相关性。【结果】 (1)高碘可引起细胞周期改变诱导其细胞凋亡的发生 ;(2 )Bcl 2表达及Bcl 2 /Bax、Bax/Bcl 2比值与高碘诱导的脑细胞凋亡有相关性 ;(3)高碘可引起大脑皮质、海马组织一氧化氮合酶活性及一氧化氮含量降低。【结论】高碘可能通过引起大脑皮质、海马细胞周期的改变 ,Bcl 2、Bax基因表达及比值改变或一氧化氮合酶活性及一氧化氮含量降低而诱导脑细胞凋亡的发生。 To investigate the mechanism of the a poptosis induced in brain by excess iodine.An animal model of goiter was reconstructed in guinea pig with feeding them water containing high level of iodine for 180 days.The effect of excess iodine on apoptosis and cell cycle in cortex and hippocamps was detec ted by using by flow cytometry;the expression of Bcl-2 and Bax was detected by i mmunofluorescent techniques;NOS activity and NO content were assayed with spectr ophotography.(1)Excess iodine may affect cell cycle and i nduce apoptosis in brain.(2)The expression of Bcl-2 and the ratio of Bcl-2/Bax ,B ax/Bcl-2 in cortex and hippocamps has correlation with the apoptosis rate.(3)Co m paried with the suitable group,the NOS activity and NO content in cortex and hip pocamps was decreased.[Conclusion]By changing cell cycle and the expression of Bcl-2 an d the ratio of Bcl-2/Bax,excess iodine may apoptosis in brain,furthemore NOS act ivity and NO content may contribute to the apoptosis.
