摘要
目的 应用膜片钳全细胞记录技术 ,观察急性分离大鼠额叶皮质Ⅴ、Ⅵ层锥体细胞自发性电活动的特点及其影响因素。方法 采用酶加机械分离法分离 8~ 12d鼠龄的Wistar大鼠额叶皮质Ⅴ、Ⅵ层锥体细胞 ,用膜片钳全细胞记录技术测定电生理学特性 ,观察不同膜电位水平时自发性电活动的特点及谷氨酸和GABA对其影响。结果 73 44 %的分离神经元具有自发性电活动 (n =64 ) ,膜电位在 -4 0~ -60mV时较易出现。 1mmol/L的谷氨酸显著增加放电活动的频率 ,10 0 μmol/LGABA则明显抑制放电活动的产生。 结论 急性分离的大鼠额叶皮质锥体细胞有自发性电活动 。
Objective To investigate the characteristics and factors influencing the spontaneous activity of acutely dissociated pyramidal cells of lays of Ⅴ and Ⅵ of the frontal cortex of Wistar rats Methods Pyramidal cells were dissociated by a combination of mechanical and enzymatic method from the brain from 8 to 12 day old Wistar rats Basic electrophysiology and spontaneous activities under different membrane potential and the influence of glutamate/GABA on the activities were studied based on whole cell patch clamp Results As many as 73 44% of the isolated neurons ( n =64) had spontaneous activities mostly appearing between a membrane potential of -40~ -60 mV The firing frequency was greatly increased by 1 mmol/L glutamate and completely inhibited by 100 μmol/L GABA Conclusion Spontaneous activities are recorded from acutely dissociated FCX pyramidal cells, which can be modulated by glutamate and GABA
