急性热应激条件下肉鸡组织器官中HSP70表达定位 被引量:3

摘要 通过观察热应激条件下肉鸡各组织中的HSP70阳性表达部位分布特点,探讨HSP70对不同组织器官的保护作用。分别在肉鸡40日龄时(选择体重差异不显著健康鸡)进行急性热应激处理,温度处理如下33℃±0.5℃、35℃±0.5℃、38℃±0.5℃各30min,修复2h分别取各处理组鸡只的法式囊、胸腺、心脏、肝脏作HSP70的免疫组织化学染色。各组织的HSP70表达时间及强度为(由强到弱):肝脏、心脏、胸腺、法氏囊。结果表明HSP70具有同源性,抗兔、抗鼠的HSP抗体可以抗肉鸡的抗原。HSP70在各组织器官中的表达定位有时间差异,不同的细胞具有不同的反应性,反应面积由大到小:肝脏、心脏、法氏囊、胸腺。HSP70的定位:肝脏、心脏定位在细胞质中。法式囊、胸腺定位在细胞质与细胞核中。 Through observing the variation of heat shock protein70 in the cell of broiler's liver? heart? thymus and IBD under the condition of heart shock, this paper discusses the effection of heat shock on theromotolerance and immunity ability. Selecting the 40 d's broilers which were unsignificance on the weight variation, broilers were subjected to 33℃±1℃?35℃±1℃?38℃±1℃ 30 min and restorated for 2 h;.Measuring the heat shock70's location by Immunohistochemistry. We found that HSP70's different express region is in different tissue (from strength to weak): liver? heart? thymus and IBD. HSP70 are located in cytoplasmic matrix of liver and heart also ytoplasmic matrix and nuclear of thymus and IBD.
