摘要 目的 :旨在研究汉坦病毒 (Hantavirus,HTV)感染非洲绿猴肾上皮 (Vero E6 )细胞后应激反应的规律及意义。 方法 :采用免疫细胞化学染色法、免疫荧光激光共聚焦及RT PCR方法观察热体克蛋白 (HSP) 70的表达规律。 结果 :HTV感染Vero E6细胞后 ,免疫细胞化学染色法可检出HSP 70阳性信号。感染后不同时间点 ,免疫荧光激光共聚焦显微镜及RT PCR观察结果表明 ,HSP 70及HSP 70mRNA的水平在感染后迅速升高 ,并持续高表达 (P <0 .0 5 )。 结论 :HTV可诱导Vero E6细胞HSP 70高表达 ;HSP Objective:To study the law and significance of stress response after Vero-E6 cells infected with Hantavirus (HTV). Methods:The law of expression of HSP 70 and its mRNA level were detected by immunocytochemical staining, laser confocal microscope and RT-PCR. Results:After being infected with HTV, immunocytochemical staining exhibited that the expression of HSP 70 increased. laser confocal microscope and RT-PCR detection showed that the level of HSP 70 and its mRNA increased quickly and kept at a high level in comparison with the normal control(P<0.05). Conclusion:The over-expression of HSP 70 in Vero-E6 can be induced by Hantavirus, which may play a role in the inhibition of virus reproduction and the protection of cells from virus infection.
