抗原抗体介导的细胞融合制备单克隆抗体

摘要 用双功能连接剂SPDP将抗原挂到SP2/0细胞表面,将该细胞与免疫小鼠脾细胞一起温育,两者借抗原抗体的特异结合而连接起来,然后按常规方法加PEG融合。我们共进行了11次这种特异性细胞融合试验,通过对得到的杂交瘤细胞分泌特异抗体的检测表明;阳性率平均为21.2%,与相同条件下进行的常规融合方法比较,阳性率提高了5.7倍。 Antigen (a new stomach tumor associated antigen) was incoporated into the membrane of myeloma cells utillizing a heterobifunctional regent SPDP. The myeloma cells coated by antigen were incubated with spleen cells from immunized mice, in this stage the myeloma cells selectively binded to antigen-reactive B-cells with the interposing antigen as a bridging ligand between the two cells. Then cell fusion was accomplished by using PEG as fusion agent. After 11 of these antigen-antibody mediated cell fusions,the result showed that 21. 2% of hybrids secreted specific antibodies. This is 5. 7 times higher than that we have observed in conventional cell fusion in our laboratory.
