粉防己多糖的提取工艺优选

摘要 目的 :利用正交试验 ,研究粉防己多糖的最佳提取条件。方法 :首先采用L8(4× 2 4 )正交试验表 ,以单糖和寡糖 (寡糖的含量以单糖表示 )的总含量为指标 ,寻找单糖和寡糖的最佳提取条件。其次用提尽单糖和寡糖的残渣以水作溶剂提取多糖。按L9(34 )正交试验表安排提取时间 ,提取次数 ,提取温度和溶媒倍数四个因素 ,均为 3个水平。结果 :提取单糖和寡糖的最佳条件为 :用 2 0倍溶媒 (6 0 %乙醇 )浸泡 2 4h后 ,于 5 0℃提取 4h ,共提取 3次 ,每次提取用2 0倍溶媒。实验证明 ,按此条件提取 ,已经提尽单糖和寡糖。多糖的最佳提取条件为 :提尽单糖和寡糖的残渣用 2 0倍溶媒 (水 )浸泡 2 4h后 ,于 10 0℃提取 3h ,重复提取一次。方差分析显示 ,4个因素均有显著性差异 (P <0 .0 1)。结论 :利用提尽单糖和寡糖的残渣研究多糖的提取条件 ,具有药材用量少、操作简单。 Objective: To study the optimum extraction conditions of polysaccharide from Stephania tetrandra S. Moore (STSM) researched by the means of orthogonal experiment. Methods: Firstly, the orthogonal experiment L 8(4×24) was arranged to search out the optimum extraction conditions of monosaccharide and oligosaccharide before extracting polysaccharide. Secondly, polysaccharide was extracted from the dregs with solvent water. The orthogonal experiment L 9(34) was arranged with four factors, which were extraction time, extraction times, extraction temperature and solvent multiple, and were arranged into three levels respectively. Results: The optimum extraction conditions of monosaccharide and oligosaccharide were four hours extraction at 50℃ after soaking one day with 20 times of solvent and extracting three times. Our experiment indicated that monosaccharide and oligosaccharide were extracted out on the optimum extraction conditions.The optimum conditions of polysaccharide were three hours at 100℃ with 20 times of solvent (water) and extracting one time. All of the four factors were significant factors (P<0.01) through analysis of differences. Conclusion: The method which is used to extract monosaccharide and oligosaccharide and research the optimum extraction conditions of polysaccharides has many advantages, such as saving much sample, time, chemical solvents as well as operating simplification.
