Purification of Copper-zinc Superoxide Dismutase from Semen Lethospermi(SL) by Copper-chelate Affinity Chromatography

摘要 本文报道了用金属螯合层析从紫草籽中分离Cu/Zn超氧化物歧化酶的一种新方法。在金属螯合层析过程中 ,运用了pH梯度和组氨酸梯度两种洗脱技术 ,取得良好效果。Cu/Zn超氧化物歧化酶最终比活为 5 36 8.75U/mg ,纯化倍数 36 6 .75 ,得率 18.7% ,并用聚丙烯酰胺凝胶电泳法证明了其均一性。 A new method for separation of Semen Lethospermi Cu/Zn superoxide dismutase ( SL Cu/Zn-SOD),including immobilized metal affinity chromatography(IMAC),has been reported Multi elution technique was used in copper-chelate affinity chromatography,increasing and decreasing pH gradient,followed by gradient elution with histidine resulted in high resolution of the enzyme SL Cu/Zn-SOD appeared to be homogeneous as judged by polyacrylamide gel electrophoresis
