内生真菌EPICOCCUM NIGRUM的形态与分子鉴定(英文) 被引量:9

MORPHOLOGICAL AND MOLECULAR IDENTIFICATION OF AN ENDOPHYTIC FUNGUS EPICOCCUM NIGRUM

摘要 在调查松属植物内生真菌过程中,从植物组织中分离到45株Epicoccum nigrum。根据它们在PDA培养基生长时的总体形态特征,划分为三组形态类型。从每组类型中随机选取三个代表菌株作进一步的分子鉴定。rDNA的ITS和5.8S基因序列分析结果表明,这9个菌株为E. nigrum。我们的研究结果也证明了前人提出的在Epicoccum菌种鉴定中,孢子大小和菌落颜色不能作为可靠的种级分类特征。 In a survey of endophytic fungi from Pinus in China, a total of 45 Epicoccum nigrum isolates were recovered. These cultures were divided into three groups based on the gross morphological characteristics on PDA. Three representative strains were randomly selected from each group for further molecular identification. The 5.8S gene and flanking internal transcribed spacer (ITS1 & 2) regions of nrDNA were amplified and sequenced. The phylogenetic relationship among the nine strains and reference Epicoccum species has been assessed by means of the ITS and 5.8S gene sequences. The analysis of DNA sequences suggests that the nine strains are identified as E. nigrum. Our result supported previous conclusion that the size of conidia and colony color could not be used as dependable taxonomic characteristics in the identification of E. nigrum.
