摘要
目的 :研究砷对淋巴细胞增殖的影响 ,观察硒对砷的拮抗作用。方法 :不同浓度的砷及砷硒联合作用淋巴细胞 ,培养 6d ,用四氮唑 (MTT)还原法观察淋巴细胞生长情况 ;用倒置显微镜观察细胞形态学改变。结果 :NaAsO2 在一定浓度范围内以浓度依赖方式抑制淋巴细胞生长 ,其IC50 为 8 6 3μmol/L ;淋巴细胞的生存率随着NaAsO2 浓度的增加而降低 ;硒的浓度为 0 5 μmol/L对砷有一定的拮抗作用。结论 :NaAsO2 对淋巴细胞的生长有抑制作用 ,且硒能够拮抗砷的细胞毒性。
Objective:To reseaech the inhibitory effects of ars en ic on lymphocyte proliferation and the antagonism effects of selenium. Methods l ymphocyte were incubated with sodium arsenite(NsO 2) alone or in combination with sodium selenite(Na 2SeO 3) for six days.MTT reductiong assay, morphology investigationg. Results:NsO 2 inhibited the growth of lymphocyte in a dose-de pendent manner in a certain range of dose with a IC 50 of 8.63μmol/L; the survival rate of lymphocyte were decreased after NaAsO 2 treatment. 0.5μmol/L Na 2SeO 3 reduced the inhibitory effects of arsenic on lymphocyte proliferation. Conclusions This study suggests that the lymphocyte proliferation can be inhibi ted by NaAsO 2 and can be antagonism by Na 2SeO 3.
