摘要
以金刺塔仙人掌嫩茎为外植体进行离体培养 ,茎段的最高分化率为 87%。在改良的MS(氮元素减半 ) +BAP1mg/L +IAA0 .2mg/L平均每个茎段可诱导不定芽 9~ 1 3个。在生根培养基中 ,以MS培养基 (氮元素减半 )加NAA0 .2mg/L ,根诱导率在 91 %。当苗高 3cm以上 ,根长 2cm以上时 ,开盖 2d移入温室 ( 2 0~ 2 6℃ ) ,炼苗 3d后移栽于蛭石 :河沙 :腐殖土为2 :3:5的混合基质的土壤中 ,成活率达 89%。
Take young stems of Astrophytum ornatum as ex-plant for vitro culture, the highest rate of stem differentiation is 87%. On the improved MS (N element decreased by half) with BAP1 mg/L+IAA0.2 mg/L, averagely every stem could induce adventitious bud by 9~13. On rooting medium, MS medium (N element decreased by half) plus NAA 0.2 mg/L, the rooting induced rate is 91%.When seedling height and root length are above 3 cm and over 2 cm respectively, open lid for 2 days and move it into greenhouse (20~26 ℃), and train seedlings after 3 days and transplant them into vermiculite: sand: humus soil with mixture medium soil by 2∶3∶5, and survival rate reaches 89%.
