摘要
[目的 ]研究脾切除对猪血清致大鼠肝纤维化的影响。 [方法 ]雄性Wistar大鼠 37只 ,随机分为正常组、模型组、切脾组和假手术组。采用猪血清腹腔注射复制大鼠肝纤维化模型 ,观察大鼠肝脏病理与胶原染色 ,检测大鼠肝功能和肝脏羟脯氨酸 (Hyp)水平。[结果 ]猪血清腹腔注射 12周后 ,模型组、切脾组和假手术组大鼠肝脏有纤细至较粗的完整的纤维间隔形成。与正常组大鼠相比 ,模型组、切脾组和假手术组大鼠血清丙氨酸氨基转移酶(ALT)活性明显升高 [(46 .79± 4 .76 )、(5 2 .5 0± 4 .4 9)、(5 1.90± 3.4 7)U/L ,P <0 .0 5 ],白蛋白明显下降 [(2 8.0 3±1.4 1)、(2 7.86± 1.6 6 )、(2 8.82± 1.37) g/L ,P <0 .0 1,<0 .0 5 ],Hyp明显升高 [(30 3.77± 32 .5 9)、(332 .6 2±5 4 .0 6 )、(30 0 .17± 4 8.0 5 ) μg/ g湿肝重 ,P <0 .0 1],而总蛋白和总胆红素无明显变化。与模型组相比 ,切脾组ALT活性明显高于模型组 (P <0 .0 5 ) ,Hyp与模型组相比有增高的趋势。[结论 ]切除脾脏不能阻止猪血清致大鼠肝纤维化模型的形成。
Objective] To investigate the effect of splenectomy on liver fibrosis induced by porcine serum. [Methods] Wistar rats were randomly divided into normal control group, model group, splenectomy group and sham operation group. Fibrosis model of rats was duplicated by peritoneal injection of porcine serum. Pathological and collagen stain of the liver were observed under microscopy. Liver function and hydroxyproline content in the liver tissue were determined. [Results] There were thin to thick complete fiber septa in the model group, splenectomy group and sham operation group after 12 weeks of injection of porcine serum. Compared with the normal group, serum ALT activity in rats of the model group, splenectomy group and sham operation group elevated significantly\[(46.79±4.76),(52.50 ±4.49),(51.90±3.47)U/L,P< 0.05 \];Alb content decreased markedly \[(28.03±1.41),(27.86±1.66),(28.82±1.37) g/L, P<0.01,<0.05\]; Hyp content increased significantly \[(303.77±32.59), (332.62±54.06), ( 300.17 ±48.05) μg/gwet liver, P<0.01\];there were no changes in TP and TBIL content in each group. Compared with model group, serum ALT activity in the rats of splenectomy group strengthened significantly (P<0.05) and Hyp content tended to increase without statistical significance. [Conclusion] Splenectomy can not stop the formation of liver fibrosis of rats induced by porcine serum.
