摘要 探讨用于DNA提取的油茶叶片保存方法,比较了冷冻(-70℃)保存与3种不同配方溶液(室温)保存油茶叶片对提取DNA质量的影响。结果表明,冷冻法保存10d、20d、30d、60d的油茶叶片提取的DNA分子量约为48kb,A260/A280值为1.833~1.936,得率为307~382μg/g(FW),与从新鲜叶片提取的DNA质量基本一致。实验结果表明,冷冻法适合于油茶叶片的保存,而3种不同配方溶液保存法不适合于油茶叶片的保存。 A comparative study on four methods for preservation of leaves of Camellia oleifera for DNA Extraction was conducted. The result indicated that the molecular weight of the extracted DNA from the leaves preserved in -70 ℃ freezer for 10 d, 20 d, 30 d and 60 d was about 48 kb,and the DNA yield was 307~382μg/g(fresh weight). The ratio of A_(260) to A_(280)of DNA was 1.833~1.936. This result was similar to that from fresh leaves. It is suggested that freezing preservation is an ideal method for leaves of C. oleifera. And other three methods, with which leaves were preserved in medium consisting of different components are not encouraging.
