摘要
目的:探讨活血通腑方对大鼠腹腔粘连转化生长因子-βmRNA表达水平的影响及与粘连级别的关系,研究其防治腹腔粘连的作用机制。方法:造模大鼠随机分为A(假手术组),B(模型组),C(活血通腑方组),D(对照组),分别进行胃内药物灌注,用逆转录-聚合酶链反应(一步法RT-PCR),凝胶图像系统分析TGF-βmRNA表达情况、非参数检验方法分析粘连变化情况,观察药物的影响作用。结果:5、21天各组光密度比值分别为0.026、0.77、0.15、0.36;0.062、0.186、0.097、0.693。其表达强度表现均为D>B>C>A,B组降低实验性大鼠TGF-βmRNA表达作用十分明显。腹腔粘连4组比较有极显著差异(P<0.01)。结论:活血通腑方防治大鼠术后腹腔粘连作用可能与其抑制TGF-βmRNA表达有关;活血通腑方能下调实验性腹腔粘连TGF-βmRNA表达水平,影响粘连程度。
objective: Probing into and invigorating blood circulation and dredging the intestines internal organs prescription(IDO Prescription) will transform growth factor - βmRNA influence of expressing competence and relation with rank of adhesion to the rat's abdominal cavity's adhesion, study its function mechanism of preventing and curing abdominal cavity's adhesion. Methods: Make rats of mould to divide for A (false operation group) at random, B (model group), C (IDO Prescription group), D (contrast the group), carry on in the stomach separately the medicine is poured into, duplicate by going against- get the enzyme chain together and react (a footwork RT- PCR ), get picture network analysis TGF - βmRNA expresses the situation, the method of inspection of the parameter analyses the adhesion change, observe the influence function of the medicine. Results : 5, The mere density ratio of every group is in 21 days respectively0.026,0.77, 0.15, 0.36,0.062,0.186,0.097,0.693. It isD>B>C>A that it expresses intensity and displays, very obvious that group B reduces experimental rat TGF- βmRNA expresses function. 4 groups of abdominal cavity' s adhesion relatively have a remarkable difference( P < 0.01). Conclusion : May be instead of suppressing TGF - βmRNA to express and have something to do with that abdominal cavity's adhesion acts on to IDO Prescription after preventing and curing the rat's skill;IDO Prescription and can reduce experimental abdominal cavity adhesion TGF- βmRNA express competence, influence the adhesion degree.
