肝力克对肝纤维化大鼠肝组织TGF-β_1PDGF-BmRNA表达的影响 被引量:3

Effects of Ganlike on the Expression of TGF - β_1 and PDGF - B mRNA in Rats with Experimental Liver Fibrosis

摘要 目的:观察中药复方肝力克对实验性肝纤维化大鼠的治疗作用及对其肝组织转化生长因子-β1(TGF-β1)和血小板源性生长因子-B(PDGF-B)mRNA表达的影响,探讨肝力克抗肝纤维化作用的可能机理。方法:138只SD大鼠随机分成正常对照组和造模组,造模组大鼠予40%CCl4 0.3ml/100g体重皮下注射;连续9周,建立大鼠纤维化模型;将肝纤维化大鼠随机分为肝纤维化模型组、肝力克大剂量组、肝力克中剂量组和肝力克小剂量组。肝力克大、中和小剂量组分别予不同剂量的中药复方肝力克灌胃治疗,肝纤维化模型组和正常对照组给予等容量的生理盐水灌胃,每天1次,连续9周。取肝组织常规HE染色,观察大鼠肝组织炎症活动度和肝纤维化程度,用原位杂交法检测大鼠肝组织中TGF-β1和PDGF-B mRNA的表达。结果:1)肝纤维化模型组大鼠肝组织炎症活动度计分及肝纤维化程度计分均较正常对照组明显升高(P<0.01);经肝力克大、中、小剂量治疗后,大鼠肝组织炎症活动度计分及肝纤维化程度计分均较肝纤维化模型组大鼠明显降低(P<0.05,P<0.01);2)肝纤维化模型组大鼠肝组织TGF-β1和PDGF-B mR-NA阳性表达均较正常对照组明显升高(P<0.01);经肝力克大、中、小剂量治疗后,大鼠肝组TGF-β1和PDGF-B mRNA阳性表达均较肝纤维化模型组明显降低(P<0.01,P<0.05);3) Objective : To study the treatment effect of Ganlike, a kind of Chinese herbs compound, on rats with experimental liver fibrosis and the influence on the experssion of TGF - β1 and PDGF - B mRNA, to explore the possible mechanism of Ganlike's and - liver - fibrotic action. Methods :138 SD rats were randomly divded into two groups:the normal control and the model groups. The hepatic fibrosis model was induced in the experimental rats by subscutaneous injection of 40% CCl4 3ml/kg/d body weight. After hepatic fibrosis model being built, the rats in the model group were divided into the hepatic fibrosis model, the high, the medium, and the low dosage Ganlike groups. Experiment disposal: The rats in the Ganlike groups were respectively given Ganlike in the different dosages, the rats in the normal control and the hepatic fibrosis model group were given the same volume of saline. At the end of the eighteenth week, all the rats were killed. The severity of inflammation and fibrosis in all liver tissues were measured by semiquantitative scoring. At last, the expression of TGF - β1 and PDGF - B mRNA in all liver tissues was deleted by in situ hybridiztion method. Results :① Compared with the normal control group, the severity of inflammation scoring and hepatic fibrosis scoring in the liver tissues of hepatic fibrosis model group was showed significant difference( P <0.01);Compared with the hepatic fibrosis model group, the severity of inflammation scoring and the liver fibrosis scoring in the liver tissues of all the Ganlike groups became obviously lower. ②The expression of TGF - β1 and PDGF - B mRNA in liver tissues of the liver - fibrotic model group was significantly higher than that of the normal group ( P < 0. 01) ; The expression of TGF - β1 and PDGF - B mRNA in rat liver tissues of all the treatment groups was obviously lower than that of the liver- fibrotic model group. ③ There was a positive correlation between the liver fibrosis scoring and the expression of TGF- β1 mRNA and PDGF- B mRNA. Conclusion ①Ganlike can relieve of the severity of inflammation and liver fibrosis in rat liver tissues. ②TGF - β1 and PDGF - B participate in the occurrence and development in hepatic fibrosis. ③ Ganlike can decrease the experession of TGF - β1 and PDGF - B mRNA, which might be a mechanism of the Ganlike ami - liver- fibrotic action.
