摘要
目的:探讨乙酰肝素酶反义寡核苷酸对人胃癌细胞株SGC7901凋亡的相关基因Fas和FasL影响.方法:胃癌细胞株SGC7901细胞由山东医科院提供.采用脂质体介导乙酰肝素酶反义寡核苷酸转染人胃癌细胞株SGC7901细胞,以逆转录-聚合酶链反应(RT-PCR)检测转染后乙酰肝素酶mRNA、FasmRNA和FasLmRNA表达的变化,采用流式细胞仪检测细胞凋亡的变化.结果:脂喷体介导转染乙酰肝素酶反义寡核苷酸后,SGC7901胃癌细胞内乙酰肝素酶mRNA和FasLmRNA表达下降(分别为1.13±0.05vs0.36±0.22,t=8.37,P=0.0001<001;1.11±0.24vs0.35±0.23,t=5.48,P=0.0003<0.01),FasmRNA表达上升(0.45±0.23vs1.02±0.02,t=6.05,P=0.0001<001),转染前后各组细胞凋亡率分别为4.04%,3.85%,3.38%,9.13%,16.00%,15.63%及13.32%.
AIM: To evaluate the effect of heparanase antisense oligodeoxynucleotide (AS-ODN) on the expression of Fas and FasL in human gastric adenocarcinoma cell line SGC7901. METHODS: Human gastric adenocarcinoma cell line SGC7901 was transTected with liposome-mediated heparanase AS-ODN. The expression of heparanase mRNA, Fas mRNA and FasL mRNA was detected after transfection using semi-quantitative RT-PCR. The apoptotic rates of transfected SGC7901 cells were detected by flow cytometry. RESULTS: The expression of heparanase and FasL gene in SGC7901 cells transfected with AS-ODN significantly decreased as compared with those in the controls (1.13 ± 0.05 vs50.36±022, t=8.37, P=0.0 001<0.01; 1.11 ± 0.24 vs0.35 ± 0.23, t =5.48, P=0.0 003<0.01 respectively), but Fas gene expression was significantly increased as compared with that in the controls (0.45 ± 0.23 vs 1.02 ± 0.02, t =6.05, P=0.0 001<0.01). The apoptotic rates were 9.13%, 16.00%, 15.63% and 13.32% in SGC7901 cells transfected with AS-ODN at the concentrations of 0.1, 0.2, 0.4 and 0.8 μmol/L, respectively. CONCLUSION: Apoptosis of human gastric carcinoma cell line SGC7901 can be effectively induced after transfected with heparanase AS-ODN.
