摘要 人们在反相色谱(RPC)和疏水作用色谱(HiC)上研究温度对蛋白质保留值影响时,常把蛋白质保留时间随温度增加而增长作为属于HIC的一个标准,将保留时间随温度增加而减小作为RPC的一个特点。实际上常常出现与之相反的保留行为。过去对于这些反常的行为解释并不令人满意。 This paper deals with the effect of temperature on retention behavior of proteins in RPC and HIC. The normal and abnormal retention behavior of protein with temperature is described. The variation in retention value is related with the extent of confor-mational change of protein and the viscosity of mobile phase in elevated temperature. As the protein conformational change makes k' large but lower viscosity makes k' small at higher temperatures, the effect of temperature on retention value is complicated depending on the two factors.
