摘要
目的 探讨 β 1,4半乳糖基转移酶 Ⅴ (β 1,4 galactosyltransferasesⅤ ,β 1,4 GalT Ⅴ )在正常和损伤脊髓组织中的定位及表达变化。方法 将 5 6只SD大鼠随机分为 7个实验组和 1个对照组 ,实验组按Allen法制作急性脊髓损伤模型 ,并分别按伤后 6、12、2 4、48、72h、7和 14d取材。将用逆转录 聚合酶链反应 (RT PCR)扩增的 β 1,4 GalT Ⅴ片段克隆到pGEM T载体中。体外转录合成地高辛标记的正、反义 β 1,4 GalT ⅤRNA探针。原位杂交分析 β 1,4 GalT ⅤmRNA在正常和损伤大鼠脊髓组织中的定位及表达变化。结果 β 1,4 GalT Ⅴ在大鼠正常脊髓灰质中表达。脊髓损伤后 12h表达明显增高 ,持续到 72h表达下降 ,但仍明显高于对照组。结论 β 1,4 GalT Ⅴ在脊髓灰质中表达 ,并在脊髓损伤后表达发生变化 ,提示 β 1,4 GalT Ⅴ在脊髓损伤中过程中起到一定作用。
Objective To investigate the locations and changes of the expression of β 1,4 galactosyltransferases Ⅴ (β 1,4 GalT) in normal and injured rat spinal cord.Methods Fifty six SD rats were divided into 7 experimental groups and one control group.The spinal cord injury (SCI) model was made according to the Allen's method.The animals were killed at different time points at 6th,12th,24th,48th,72nd h,7th day and 14th day after the injury.The expression of β 1,4 GalT Ⅴ in mouse spinal cord was detected by RT PCR.The products of RT PCR were used to label probes for in situ hybridization and the expression of β 1,4 GalT Ⅴ in injured rat spinal cord was analyzed by in situ hybridization.Results β 1,4 GalT Ⅴ mRNA localized mainly in the grey matter of spinal cord.The expression of β 1,4 GalT Ⅴ was increased significantly at 12th h after SCI,then decreased gradually at 72nd h after injury.Conclusion The change of the expression of β 1,4 GalT Ⅴ in injured spinal cord provided an evidence that β 1,4 GalT Ⅴ might be involved in the regeneration or degeneration of the injured spinal cord.
