摘要
本试验筛选出了 45个引物 ,可产生遗传多样性 ,其中OPA 0 8、OPA 16、OPC 10、OPC 12、GENMES’S 36号、78号、112号、181号引物其扩增结果稳定性好、多态性强。这些引物的RAPD扩增结果都可作为烟草烤烟品种的遗传多样性分析、基因定位及分子标记等。经凝胶电泳比较 ,GENMED’S 36号引物可作为烟草品种的最适宜的引物 ;1.8kb条带被初步认为与抗黑胫病基因相关。这些结果为以后对烟草烤烟品种在分子水平上的研究奠定了基础 。
Thirty three primers were selected from one hundred Operon Primers and another twelve Primers were selected from one hundred Genmed‘s Primers:all of them can generate genetic variation. The OPA 08、OPA 16、OPC 10、OPC 12、GENMED’S number 36、78、112、181 can produce stable and intense polymorphisms bands. All result can be used as analysis of genetic variation, gene location and molecular marker for flue cured tobacco varieties. By compared synelgel electrophoresis, GENMED'S 36 Primer can be used as apecial primer for flue cured tobacco varieties. The 1.8kb hand may be related to black shank resistant gene, accordingly, it established the base of the research of flue cured tobacco varieties and accelerated the research course of it in molecular level.
