摘要
目的 研究旋毛虫新生幼虫T6 6 8重组抗原的免疫原性 ,制备基因工程疫苗。方法 以旋毛虫新生幼虫期特异性基因T6 6 8在大肠杆菌中的表达蛋白为抗原免疫小鼠 ,每间隔 10d免疫 1次 ,共免疫 3次。末次免疫后 10d ,每只小鼠攻击感染 2 0 0条旋毛虫感染性肌幼虫 ,感染后 5周用消化法检查肌幼虫 (ML)负荷。结果 T6 6 8重组抗原免疫组肌幼虫减虫率明显高于佐剂组和对照组。结论 T6 6 8重组抗原能诱导小鼠产生一定程度的保护性免疫 ,且可激发特异性体液免疫。
Objective To study the immunogenicity of Trichinella spiralis (T.s) newborn larvae (NBL) recombinant antigen, and to produce genic engineer vaccine. Methods The NBL stage-specific gene T668 was expressed in E.coli and the expression protein was used to immunize the mice at an interval of 10 days. Ten days after the final immunization, the animals were challenged with 200 T.s infective muscle larvae (ML) per mice. ML loading was detected by digest method at week 5 after the challenge infection. Results The reduction rate of ML in immunized group was significantly higher than that in the adjuvant and control group. Conclusion T668 recombinant antigen could induce protective immunity as well as specific humoral immunity.
