β-七叶皂甙治疗创伤性脑水肿分子作用机制的实验研究

摘要 目的 研究 β-七叶皂甙 (β -Aescin)对大鼠急性脑损伤后脑组织NF -κB活性的影响。方法  6 2只SD大鼠采用自由落体致伤法制作脑外伤模型 ,随机分成假手术对照组 (A组 )、创伤组 (B组 )、β -七叶皂甙治疗组 (C组 )、吡咯烷二硫基甲酸酯 (PDTC)治疗组 (D组 ) ,每组分别在术后 6h、2 4h和 3d 3个时点收取脑组织标本 ,采用凝胶电泳迁移率分析法 (EMSA)测定脑组织NF -κB活性 ;同时测量脑组织含水量以及做病理形态学观察。结果 与A组比较 ,B组大鼠脑损伤后脑组织NF -κB活性和脑组织含水量显著增高 (P <0 .0 1) ;C、D组无显著性差异 ,但D组脑组织含水量略有增高 ;与B组比较 ,C组和D组脑组织NF -κB活性 (P <0 .0 1)、脑组织含水量 (P <0 .0 5 )均显著降低。结论 β-七叶皂甙可以抑制NF -κB活化 ,可能是 β -七叶皂甙治疗创伤性脑水肿的分子作用机制之一。 Objective It is to study the influence of β-Aescin on the activity of NF-κB in rats' brain tissue after acute brain injury.Methods 62 SD rats were made into brain injury models with free falling object vulnerant method and randomly divided into notho operation control group (group A), injury group (group B), β-Aescin treatment group (group C), PDTC treatment group (group D). The brain tissue specimen in every group was respectively gathered at 6 h, 24 h and 3 d after operation. The activity of NF-κB in brain tissue was measured with EMSA method, at the same time the moisture in brain tissue was measured and a pathomorphology observation was done.Results Compared with group A, the activity of NF-κB and moisture in brain tissue in group B were obviously increased ( P <0.01). Compared with group B, the activity of NF-κB ( P <0.01) and moisture in brain tissue ( P <0.05) in group C and group D were all obviously decreased.Conclusion β-Aescin can inhibit the activity of NF-κB. This may be one of molecular mechanisms of β-Aescin on traumatic brain edema.
