摘要
目的 :探讨鹿瓜多肽注射液治疗类风湿性关节炎 (RA)的机理。方法 :以Ⅱ型胶原 (CⅡ )诱导的免疫性关节炎 (CIA)大鼠为RA动物模型 ,通过测量其关节肿胀程度及制作病理切片并在光镜下计分 ,观察鹿瓜多肽注射液对其的治疗效果 ,并取膝关节滑膜切片进行原位杂交 ,用图像分析系统检测MMP3 和TIMP 1mRNA表达水平。结果 :与模型组比较 ,鹿瓜多肽组大鼠踝关节肿胀程度和病理损伤计分均明显减轻。模型组大鼠滑膜组织MMP3 mRNA表达阳性密度明显高于正常对照组 ,而TIMP 1mRNA表达阳性密度明显低于正常对照组。鹿瓜多肽组大鼠滑膜组织MMP3 表达降低 ,TIMP 1表达增高。结论 :鹿瓜多肽注射液对CⅡ诱导的免疫性关节炎有治疗作用 ,其机理可能与抑制滑膜组织MMP3 mRNA表达并促进TIMP 1mRNA表达有关。
Objective:To probe into the mechanism of Lugua polypeptide injection on immune arthritis induced by type Ⅱ collagen.Method:Arthritis of rat induced by type Ⅱ collagen was used as rheumatoid arthritis model.Selling of ankles of rats were measured and pathological sections were observed and scored to assessed the therapeutic effect.The level of MMP 3/TIMP 1 in synovium was investigation with hybridization in situ.Result:Compared with that of model group,Swelling of Lugua polypeptide group was relieved significantly.Significant increase of MMP 3 mRNA expression and decrease of TIMP 1 mRNA expression can be observed in synovium of model group.But in Lugua polypeptide group,the increased expression of MMP 3 mRNA was lowered and the decreased expression of TIMP 1 mRNA highered was observed.Conclusion:Inhibitio nof MMP 3 mRNA expression and promotion of TIMP 1 mRNA expresison maybe one of the mechanisms of Lugua polypeptides effect on immune arthritis induced by type Ⅱ collagen.
