摘要
目的研究苯暴露对苯作业工人外周血细胞DNA断裂损伤的影响,并进一步探讨苯暴露与DNA断裂损伤间的剂量-反应关系。方法用3M有机气体被动式采样器监测个体接苯浓度,计算8小时时间加权平均浓度(8hTWA)和累积接苯浓度。用单细胞凝胶电泳试验检测苯作业工人外周血细胞DNA损伤,观察指标包括Olive尾矩和DNA断裂分级。结果接苯工人(51人)Olive尾矩和DNA断裂分级两个参数,均明显高于对照组(31人)(F=3003,P<00001;χ2=2399,P<00001),并均与TWA浓度和累积浓度有剂量-反应关系。Olive尾矩与TWA浓度和累积浓度显著正相关。结论苯暴露导致外周血细胞DNA断裂损伤增加,且呈明显剂量-反应关系,累积接苯浓度比单纯苯浓度更能反映苯暴露水平。
Objective To investigate the effect of benzene exposure on DNA damage of the peripheral white blood cells and to assess the possible dose-response relationship between benzene and DNA damage. Methods Personal benzene exposure was sampled with 3M organic vapor monitors. The time weighted average concentration (8h-TWA) and the cumulative dose were calculated. Single cell gel electrophoresis assay was used to detect DNA damage in white blood cells of benzene-exposed workers. The Olive tail moment and the grade of DNA breakage were used to measure DNA damage. Results The Olive tail moment and the grade of DNA breakage in benzene exposure groups were significantly increased in comparison with those in the control group (F=30.03,P<0.0001,χ2=239.9, P<0.0001,respectively) and showed a dose-response relationship with benzene concentration. Correlation analysis showed that Olive tail moment was correlated with benzene exposure concentration. Conclusions Benzene exposure resulted in an increase of DNA damage of the peripheral white blood cell, which was in a dose-response relationship manner; cumulative dose was better than simply concentration to reflect benzene exposure.
