摘要
研究了异宗结合食用菌的原生质体单核化现象和单核原生质体的遗传规律。结果表明,在原生质体制备和再生过程中形成单核原生质体,是异宗结合食用菌的一个普遍现象。在“佛罗里达”(香菇)等异宗结合食用菌的原生质体释放中,单核原生质体和双核原生质体之比由0.85∶1到2.34∶1不等。经测定。这些单核原生质体具有亲本双核菌株中存在的两种交配型(AxBx,AyBy),但两种交配型的比例有所不同。单核原生质体再生菌丝的菌落形态、菌丝生长速度和营养缺陷型标记等性状均稳定地保持了亲本的特性。由于这种单核体与孢子萌发形成的单核体,具有不同的遗传背景,故利用它作为杂交育种的亲本,可以克服食用菌常规育种中的很多局限性,具有广阔的应用前景。
Monokaryotization by protoplasting and genetic rule of protoplasted monokaryon in heterokaryotie mushrooms were investigated.Appearing of protoplasted monokaryon was a general phenomenon in release and regeneration of protoplast from mycelia of mushroom.The ratio between dikaryon and monokaryon was 0.85 and 2.34:1 in different strains of mushrooms.There were the same mating types of parents(AxBx,AyBy) in these protoplasted monokaryons,but the ratio between the two mating types varied in dif- ferent strains.The characters of parents,such as morphology of colony,growth rate of mycelium and auxotroph,were maintained in protoplasted monokaryon.There was differ- ence in genetic background between protoplasted monokaryon and sporulated monokaryon. Protoplasted monokaryon was asexual progeny without miosis and sporulated monokaryon was sexual progeny by miosis.
