摘要
目的研究干扰素α-2b对H22鼠肝癌细胞株体内外的抑制作用。方法MTT法测定不同浓度干扰素α-2b对H22鼠肝癌细胞的作用。建立小鼠皮下实体瘤模型,腹腔内分别注射生理盐水、干扰素α-2b、氟尿嘧啶(5-FU)、干扰素α-2b+5-FU,隔天1次,共给药10次,停药1周后处死动物,测量瘤体大小、瘤重,计算抑瘤率,测定微血管密度(MVD)。结果干扰素α-2b对体外培养的H22鼠肝癌细胞无直接抑制作用,但明显抑制实体瘤生长;干扰素α-2b、5-FU、干扰素α-2b+5-FU抑瘤率分别为49.5%、67.4%、74.9%,MVD分别为67±43、134±53、6.3±46。结论干扰素α-2b具有抗血管生成作用,与化疗药物5-FU联用有协同作用。
Objective To study the effect of Interferon α-2b on the growth of H_~22 murine hepatocellar carcinoma cells in vitro and in vivo.?Methods The effect of Interferon α-2b on murine H_~22 cells was investigated in vitro by using MTT colorimetric assay. Mouse model of murine hepatocellar carcinoma was established by implanting H_~22 murine hepatocellar carcinoma cells subcutaneously. The model mice were randomly divided into 4 groups:control,5-Fu,Interferon and Interferon+5-Fu (with drugs given i.p. qod×10 times,respectively). One week after the completion of drug administration,the mice were killed to evaluate the tumor volume, tumor weight, inhibition rate and intratumor microvessel density (MVD).?Results The MTT colorimetric assay showed there were no direct effects of Interferon α-2b on the murine H_~22 cell line. The in vivo experiment showed the tumors in the mice treated with Interferon α-2b,5-Fu or Interferon α-2b+5-Fu,grew at a slowed rate with the inhibition rates being 67.4%,49.5% and 74.9%,respectively,and MVD being (13.4±5.3),(6.7±4.3) and (6.3±4.6),respectively.?Conclusion Interferon α-2b can inhibit tumor growth by inhibition of angiogenesis,rather than by antiproliferation. Interferon α-2b and 5-Fu are synergetic in offering this effect.
